Next Generation Sequencing (NGS)
Next-generation sequencing (NGS) is a collective term for sequencing methods that utilize parallel reading of thousands to millions of nucleic acid fragments in a single sequencing run. As a result, these technologies enable the generation of significantly more comprehensive and information-rich data compared to targeted alternative methods.
Among the most widely used approaches in the field of NGS is Sequencing by Synthesis (SBS), on which Illumina platforms are based. Our portfolio includes NGS kits compatible with Illumina sequencers for research use in oncology and pharmacogenetics, specifically the analysis of the BCHE gene in connection with the prediction of adverse reactions to certain anesthetics. The products are intended exclusively for research use (Research Use Only, RUO) and are not intended for clinical diagnostics.
BCHEseq
Examination of the BCHE gene encoding the enzyme butyrylcholinesterase is especially important in anesthesiology. Several variants in the gene have been described that cause enzyme deficiency, leading to impaired metabolism of the anesthetics succinylcholine and mivacurium. A decrease of BCHE activity may result in complications such as prolonged muscle paralysis after awakening from anesthesia following the use of these anesthetics, potentially requiring respiratory support, even mechanical ventilation. The kit allows sequencing of the complete coding sequence of the BCHE gene. This makes it possible to detect the full spectrum of SNPs and microdeletions/insertions in the coding region of the gene, including the clinically most significant mutations A, K, F1, F2, and S.
Kit parameters
Contains reagents for processing 32 samples with unique index combinations into one sequencing run on the Illumina MiSeq instrument.
The starting material is DNA isolated from patient blood.
The recommended input amount of DNA per sample is 10–20 ng.
The total library preparation time (until sequencing begins) is approximately 5 hours.
The library is compatible with the sequencing primers included in the kit.
The kit is intended for research and scientific purposes only, not for clinical diagnostics. „Research-use only“
Created with financial support from the Technology Agency of the Czech Republic (TAČR).
NGS MSI-test
The BRAF, KRAS, NRAS genes and microsatellite instability (MSI) are important especially in oncological diagnostics for determination of the suitability of a particular biological therapy and disease prognosis, and also serve as a clue in differential diagnostics when clarifying the possible hereditary origin of the disease. The kit contains reagents for preparing an NGS library for sequencing on Illumina MiSeq platforms using the amplicon sequencing method (multiplex PCR). The library covers five microsatellite markers (BAT25, BAT26, NR21, NR22, NR24) and hot-spot regions of the genes BRAF (exons 11, 15), KRAS (exons 2, 3, 4) and NRAS (exons 2, 3, 4).
